Method overview

Common microbiome methods include amplicon sequencing (16S, ITS and 18S) and metagenomic sequencing. The original description notes limits of conventional 16S profiling for species-level identification and functional interpretation, and the sequencing and computational costs of metagenomics. It describes SMURF/5R, a multiplex PCR approach covering five 16S hypervariable regions, as a route to species-level resolution. According to the source, its amplified regions cover approximately 68% of the full-length 16S sequence and the method has appeared in more than 30 international publications, including Cell, Science and Nature Communications.
