Method overview

Particle-size testing is an important part of EV research, providing information on size and particle-number distributions and purity for downstream functional studies and analysis. The source describes particle-size analysis as a rapid, reproducible complement to other characterization methods.

The laboratory uses the Flow NanoAnalyzer from NanoFCM and the ZetaView nanoparticle tracking analyzer from Particle Metrix, Germany, for EV characterization. The instruments are described below:

1. Nano-flow cytometry (NanoFCM)

The Flow NanoAnalyzer covers particle sizes below 200 nm, a range described in the source as a blind spot for conventional flow cytometry. It characterizes nanoparticles and natural biological nanoparticles including subcellular structures, bacteria, viruses and extracellular vesicles. The source describes high-resolution, selective and high-throughput analysis of individual nanoparticles (7–1,000 nm), including size distribution, concentration and biochemical properties.

2. Nanoparticle tracking analysis (NTA)

The source describes ZetaView single-particle tracking in combination with microelectrophoresis and Brownian motion analysis. Automatic calibration, autofocus and scanning of sample subvolumes are used to calculate size and zeta-potential distributions from thousands of particles. Particle concentration can be estimated by video counting; translational diffusion is calculated from Brownian motion, and electrophoretic mobility is used to derive zeta potential.
