Method overview
EV isolation options include ultracentrifugation (UC), polymer precipitation, tangential flow filtration (TFF), ultrafiltration (UF), size-exclusion chromatography (SEC) and affinity capture. Select a workflow according to sample type, volume, target fraction and downstream experiment, balancing recovery, purity and cost.
The laboratory has ultracentrifuges, automated EV isolation instruments, TFF systems and SEC columns. Existing service configurations cover supernatant inputs from 100 µL to 100 L, subject to sample and workflow assessment. Downstream options include characterization, purity and contaminant checks, and lyophilization.

1. Ultracentrifugation(UC)
Differential ultracentrifugation removes cells, debris and larger particles before enriching EVs, with washing as required. Sucrose cushions or iodixanol gradients can add separation steps to reduce some co-isolated components. Purity and recovery depend on the sample and protocol and must be assessed by characterization and contaminant checks.

2. TFF concentration and SEC purification
TFF directs flow along a membrane for EV concentration and buffer exchange. SEC separates particles by access to pores in its packing material: larger EVs generally elute earlier, while smaller soluble proteins elute later. TFF and SEC can be combined for larger inputs; recovery, purity, processing time and cost require sample-specific evaluation.

3. Automated EV isolation (EXODUS)
EXODUS combines negative-pressure oscillation with double-coupled harmonic oscillation to drive membrane vibration. A nanoporous membrane enriches EVs while some permeable components pass through. The technology was reported in Nature Methods; recovery, integrity and residual contamination still require measurement for each sample type.

4. Automated EV Membrane-sensing peptide capture (EXO-Smart)
Membrane-sensing peptides interact with EV membranes through features such as curvature, lipid composition and lipid-packing defects. Selected peptides can be coupled to magnetic beads for automated capture and elution. The recovered subpopulations and efficiency depend on peptide design and sample conditions and should be evaluated with controls and characterization.
