Method overview
Extracellular vesicles contain RNA types such as mRNA, miRNA and lncRNA. Sequencing these RNAs provides information for studies of EV RNA and its functions. EV RNA sequencing has been used in research on cancer, neurodegenerative and cardiovascular diseases, including investigations of potential biomarkers. The service typically includes RNA extraction, sequencing and data analysis. Depending on the assay and analysis, results may support studies of RNA expression, splice variation and post-transcriptional modifications.
1. Small RNASequencing
Small RNAs include miRNA, piRNA, tsRNA and snoRNA, and participate in gene regulation, RNA processing, translation, responses to genetic invasion and gamete formation. miRNAs have been associated with diseases including cancer, cardiovascular disease and immune disorders. EV miRNAs are also investigated as potential biomarkers. High-throughput sequencing can profile small RNAs and compare their expression across disease states, sample sources or treatment conditions to guide further research.
The source describes a QIAseq-based library preparation workflow with a stated minimum input of 1 ng total RNA. It uses optimized reaction chemistry to reduce library preparation bias and adapter dimers, and unique molecular identifiers (UMIs) to label miRNAs early in the workflow. Input suitability and data quality depend on the sample and should be assessed for each project.

2. Non-coding RNA sequencing
EV RNA, particularly non-coding RNA, can show differences in expression across disease models and is being investigated for liquid biopsy and disease mechanisms. High-throughput transcriptome sequencing can detect RNA molecules such as lncRNA, mRNA and circRNA to support studies of EV RNA identity and regulatory function.
The source describes a SMARTer-based library workflow with a stated minimum input of 1 ng total RNA for lncRNA library preparation. Sequencing data can be analyzed for lncRNA and mRNA separately or combined with miRNA sequencing data for a broader transcriptome analysis including lncRNA, mRNA, circRNA and miRNA.

Lifeint provides EV research and application services from sample preparation, isolation and characterization to omics, functional studies, modification and scale-up. It jointly established a biomedical technology centre with Xiamen University School of Pharmaceutical Sciences, built process-development laboratories to GMP standards, and contributed to an EV expert consensus. Its service records cover more than 300 plant species.
